Please I would like to know that, Is it good idea to culture Dental pulp stem cells(DPSCs) in the Alpha MEM media without L-ascorbic acid? I isolated DPSCs from mice molar teeth. Ascorbic acid is known to induce osteoblast differentiation of mesenchymal stem cells so did not want to have that in the media. Does that have any effect in proliferation of DPSCs? Actually I want to study the senescence of those cells in different conditions.
DPSCs seem to grow just fine on MEM according to the attached protocols from SciGine. However, it seems ascorbic acid is used in all associated cell culture protocols that I can find. I would imagine that it's only there for pH but I may be wrong.

Thank you very much. The cultured DPSCs without Ascorbic acid also growing well......... I am just worried about the chances of differentiation after adding ascorbic acid...
I would suggest that you do not add the L-ascorbic acid during the passaging stages. However you might have to add it to the media, if you are studying differentiation(alkaline phosphatase,alizarin red etc etc)
