Does ROS or lipid ROS increase, if detached cell is treated by drug inducing ROS, despite of adherently growing cell originally ?
Protocol of ab113851 DCFDA ROS kit recommends staining cell first and then carry out treatments (1~6 hours), followed by meaurement of ROS in the presence of compound/experimental conditions to prevent removal ROS species during wash steps.
Tough question to answer. In the case of N-acetylcysteine, low to moderate dosages promotes lipid peroration. Meanwhile moderate to high dosages act as an antioxidant. Antioxidants can become proxidants and then serve as antioxidants!
Hi, good question! I would advice to run a lot of control samples (compound untreated) to distinguish between the 'ROS background' linked to your experimental protocol from the ROS production/accumulation due to your compound treatment.
