I am looking to run westerns on cells that have grown on transwells for ~4months - do you have to pool multiple transwell inserts in order to have sufficient protein or is one transwell enough?
12-well plate is enough for WB analysis. As you know the protein conc. will very on cells type.
Briefly, After 1) Fixing by ice-cold PBS, 2) a. add 20-50uL of lysis buffer and run it thoroughly, 3) keep it dark at 4C over an hour then 4) centrifuge and collect the supernatant.
2) b. or you can collect cells into e-tube then add lysis buffer (ProPrep/RIPA)
