Hello, everyone. While transfecting my cells with plasmid, I noticed this overnight; normally, my cells are adherent (as seen in the background); I'd like to know if anyone else has seen this before; is it a contamination? I didn't see any media color changes. and this is a floating separate component, not just one. Thank you for any suggestions.
It's hard to say for sure, it could be a contamination. First I would check the medium you used under the microscope and see if something is in it. Then I would suggest preparing a completely fresh bottle of medium, sometimes additives like FCS can be the issue. Then, defreeze a new batch of cells and grow them in the fresh medium. For experiments, I wouldn't risk continuing with those cells. Also a deep clean of your cells culture especially flow hood/incubator may be in order.
Thank you Yvonne Kraus , I'd like to add also that it's might be some gelatine particles were damaged by the transfection reagent, as I coat the plates with gelatine before seeding the cells? (in my backup cell culture, nothing was visible after one week of culture).
That doesn't look like contamination, hard to say for sure. As you Rebecca Youssef mentioned, it might be some gelatin particles. Is it possible, these are dead cells since cells die upon transfection as well? But not sure whether dead cells clump together like this.
Anushka Srivastava , Thank you for your response; nevertheless, I do not believe these are dead cell clumps because I can see dead cells floating underneath. I have several hypotheses that this could be the case: 1) gelatine 2- contamination (which doesn't appear to be the case). 3. The cells had secreted something. 4- PSC (I've seen multiple publications with the same morphology that has been identified as PSC, as seen in the attached photo, but I'm not sure how much it is feasible by o/n plasmid transfection to get this)
