I am a little bit confused by the concept that priming BMDMs with LPS for Nlrc4 inflammasome studies. Originally I thought this LPS priming is essential for Nlrp3 due to it activates Nlrp3 gene transcription. And of course, it also activates IL-1B.
But does LPS priming also activates Nlrc4 gene transcription? What's the purpose of "priming" for Nlrc4?

Hello, actually most recent studies dealing with NLRC4 inflammasome use LPS in the priming phase and MDP or salmonella in the activation phase. So I think that LPS can also promote the activation of NLRC4 gene. I hope this paper can further help you: Article The NLRC4 Inflammasome
Hi Iskander Madhi , Thank you very much and this is a nice review on NLRC4.
However, the review states that the basal level of NLRC4 is sufficient to initiate NLRC4 inflammasome activation. It seems like the purpose of the priming step is not primarily inducing more Nlrc4 transcriptionally. Also, this review mentioned that Tnfa, not LPS or Pam3csk4 generally used in priming step, induces Nlrc4 transcription. So I still don't get it why people do lps priming before salmonella infection. I am guessing it's just for the induction of IL1B?
Xingchen Dong yes you are right, there is no clear evidence that LPS activates transcriptionally NLRC4. I join your guess, LPS is used for pro-IL-1B, pro-Caspase-1 and NLRP3 induction, and these proteins will help in the NLRC4 inflammasome activation. there are some studies showing that NLRP3 is involved in NLRC4 inflammasome regulation. I think you should check them to get some more informations. Good luck
