I used Cytochalasin D as an actin inhibitor in epithelial cells. Crystals formed and blocked the whole well (96-well plate) at 1 µM concentration, and formed few crystals at 0.5 µM, and no crystals at 0.25 µM and 0.1µM. It continued to inhibit cell migration even at lower concentrations. I'm just curious, why does it crystalize.

How do you prepare cytochalasin D? It is recommended that you make a stock solution in the required solvent in which it dissolves. Then dilute the stock in the appropriate aqueous medium to provide a physiologically acceptable final concentration in culture. The solvent used to dissolve cytochalasin D could be toxic to cells if it exceeds the required limit in the final concentration used in culture.

Malcolm Nobre Thank you for your answer. I got the cytochalasin D already dissolved in DMSO. and I diluted it in medium to reach desired concentration. I understand that DMSO is toxic to the cells, however, they didn't die, they just stopped migrating and looked a bit unhealthy I would say, specially at the higher concentrations I used. DMSO dissolves at 37 C, my cells were incubated, so I don't think the crystals are from the DMSO.
