3T3-L1 cells grow healthily when the humidity of the incubator is maintained by using a tray of water but it grows unhealthily and slow when the copper sulphate solution is used to maintain humidity. Does it really affect 3T3-L1? Are 3T3-L1 cells sensitive to copper sulphate?

I work with NIH-3T3 fibroblast and I use copper sulphate too. I do not have problems with the use of CuSO4 to maintain humidity. I use this method to culture others cell lines and primary culture and the results is the same, CuSO4 does not affect the growth of cells.
I think that may be there are something wrong with the medium (pH, L-glutamine concentration, serum, etc) or another component.

I've run 3T3-L1 in both their undifferentiated and differentiated state with CuSO4 in the water tray and had no issues, although this is the only way I've run them so maybe they would have done better without it there. I wouldn't of thought that you'd get much copper volatilizing and affecting the cells. Was this a one off observation or have you tried this repeatedly with the same results?
