Hi all-
I have been conducted my research for protein expression with e. coli (BL21-DE3 strain) and I usually use BLEACH for my liquid waste (usually LB waste).
And what I found is the color of liquid waste turned to dark-brown (or black) whenever my expression does not go well.
Does anyone know why this happen?
- Usually, when I use fully-grown starting culture (OD600 is above 2), liquid waste turned to dark-brown color. :-(
Maybe you used too much or too concentrated bleach and it oxidized everything to carbon.

It is because of Maillard reaction between sugar and proteins to produce Maillard products those of which are dark brown.

I believe the Maillard reaction occurs only upon addition of heat, e.g., frying (to 300 deg F); in this case, bleach was added to the liquid waste at room temperature.
