I'm really interested to know more about proteins in FBS. Recently, I ran Native PAGE with FBS and I saw a number of bands at the range between 700KDa-800KDa. Does anyone know what kind of protein that is? I tried to surf the literature, but nothing helps. Major proteins are albumin (66KDa) and globulin (too many sizes). Has anyone reported a profile in FBS/ran FBS in a gel?
FBS is going to be packed full of many types of proteins and aggregations of proteins. So the bands that you may be seeing could be a dimer, trimer or greater. Another point to note is that not all FBS will be the same, they'll vary in exact composition batch to batch and source to source. I'm pretty sure there'll be literature out there that will summarise on the major components and proteins that are in FBS but I don't think there'll be one electrophoresis profile out there that would fit all.
Thanks for the reply, Yea, I'm aware of that just today as I tried to detect my protein in my culture media. My protein is around 700-800KDa and it just came to my mind to check it with FBS. And out of all band, why do the FBS has any protein similar to my protein size. I was so frustrated and that is why I tried to find it. As far as I had found, Most of the study only describe the presence of protein lower than 200KDa and that make me wonder about. Maybe the only way to know if we can do seperate western blot against the protein I want and those from FBS.
