Thanks a lot for your helpful answer, indeed! I am going to downregulate multiple genes of a pathway in Salmonella using dCas12a. The tools developed for E. coli, typically work in Salmonella. Actually, I am using the codon-optimized catalytically inactivate (D908A) variant of AsCas12a (obtained from your recent publication https://pubs.acs.org/doi/10.1021/acssynbio.0c00424) for expression in Salmonella. I will contact CCTop team to see if they have any suggestions for Salmonella.
What about chopchop (https://chopchop.cbu.uib.no/)? Chopchop suggests a number of gRNAs for each gene with different (high to low) scores, binding upstream and N-terminal region of the genes, both forward and reverse DNA strands.
