i am planning to do ELISA for AB quantification in an Alzheimer related project. the sample is cell culture (not animal samples), since AB deposits in the extracellular environment should i use only the supernatant or the cells?
It depends on the type of your cells. Hippocampal cells (CA1) for example can internalize beta amyloid. I would recommend to homogenize your sample and use both cells and supernatant.
If it is secreted and extracellular concentration is sufficient to perform the experiment, adding lysate-originated proteins means a more complicated sample and increased vulnerability to interference. I just prefer the secretome to be analyzed if AB is completely secreted into media. You may verify it first by quantifying them separately (intracellular and extracellular).
