I am working with SureSelect XT2 all exons-library prep protocol and I am getting different yields every time I am using this protocol. I am a little uncertain about the first Ampure clean up step since we are using 180ul of Ampure xp and the elution time is only 2 minute. Is anyone facing the same issue with this protocol?

Hi Arezoo,
When you perform a purification with Ampure, it's critical the time drying your beads when you remove the ethanol in the wash step. If you over-dry your beads, the DNA won't elute properly and the recovery yield will vary a lot.
