I am trying to label LSD1 using Click chemistry. I would like to show the fluorescence by SDS Page. It worked so fare with pure protein (37ug) even though I had a lot of Background. I would like to show the same with cells. Does anyone is familiar with labelling histone proteins?

You could use a ligand to improve the efficiency of cycloaddition. You can try THPTA or other water-soluble ligand (Hong, Presolski, Finn; Angewandte Chemie Int. Ed., 2009), if you didn't try yet. Thank you very much for your replies. I am trying to label LSD1 and do competitive assays using TAMRA azide. I had quite a good result using pure protein but I do not see any signal using cells. Selva chemist, which proteins are you/did you label so far?
