I am trying to extract DNA from FFPE tissue. I tried seveal protocols using the QIAamp DNA FFPE Tissue Kit. I usually get a good yield but with a low 260/230 ratio around 0.5 I think this may be due to formaldehyde cross-linkage. What is the best way to remove formaldehyde from the tissue without affecting the DNA quality?
I think you can try to precipitate DNA with EtOH and NaCl, as indicated in Maniatis, followed by double EtOH 70% washing. Anyway,I've been working with FFPE extracted DNA for 2 years, and the A260/230 value, that you indicated, is quite normal. With that value, you can anyway perform PCR
Thank you Francesca Simonato. It appears that I have to make a blank using the ATE buffer not water on using the nanodrop spectrophotometer. Now i can get a got 260/230 ratio more than 1.7
