Ezifast like many precolumn derivitisation methods requires care in use to avoid matrix interferance. Prep requires additional care due to small volumes utilised this is why the specialist pippette is provided.
I personally have found post column derivitisation to be a more rugged technique for the determination of amino acids where matrix effect is not fully able to be explored for this work I have used nihydrine as the derivitising agent

I don't use the kit. I use a protocol that derivitizes it with ethyl chloroformate. The kit uses propyl chloroformate and n-propanol. Derivitization with methyl ethyl and isobutyl chloroformate are hot in metabolomics now. It can do every amino acid except arginine. Serine and Threonine can also be a little troublesome.
