We wolud like to target fibroblast population in our cell culture compare to our cells of interest. We already have aspecific dye to recognize our cells of interest but we would like to know the proportion of fibroblast within our cell culture plates by flow cytometry and eventually fluorescence microscopy.
Do you mean a specific marker? Especially for flow, I don't think you can just use a dye for analysis.
Can you give us a little bit more info on your experiment:
- What do you mean by dye? You can use antibodies to stain the fibroblasts.
- Are these mouse or human fibroblasts, because that will make a difference in markers? For human you can use clone TE-7 from chemicon.
- Is your cell culture a mixture of cells from for example a tissue or is it a pure fibroblast culture?
- Which technique are you planning to use for staining? FACS or immunofluorescence or both? For FACS of mouse fibroblasts you can use CD90, but you will have to include CD45 because this is also expressed on NK cells etc.
- Will you fix the cells for microscopy or will you do live imaging?
