Despite my effort, I was unable to find on the web a human cancer cell line proven to be FOXO3A-null.
Does anybody know about the existance of such a kind of cells?
For some reasons, sh-transduced cells do not fit my needs.
Also, a prostate cancer cell line would be preferable, though not essential.

Would a MEF cell-line do? I know that a FOXO3A knock-out mouse was described by Ron DePinho about 10 years ago. He is likely to have created a cell-line.
As for prostate cells- according to Genecards, PC3 cells have much lower expression of FOXO3a wheras LnCaP are high. Maybe this differential expression would be sufficient for your work?
Or, you could consider knocking the gene out of your cell line yourself using CRISPR technology, which is relatively straightforward.
Yes Amanda, I was aware of that knock-out mouse, but I need to work on human cells that completely lack FOXO3 expression.
If I can't find any FOXO3-null cell line, I will use the CRISPR technique, but as it is a pretty new technology, I would keep it as my ultimate option.
