Hello,
I've read a lot online that we are supposed to check that the pH of Towan Buffer is ~8.3 however I couldn't find whether that should be the pH of the 1x buffer after addition of methanol or before addition of methanol.
I usually add 20% methanol to my 1x of 25 mM Tris base and 192 mM Glycine. This is for a wet transfer using the Tank.

Looks to me the Towan buffer has strong suffering capacity. Adding methanol should not affect the pH value. For membrane transfer, I don't think that will be a problem

Independently on the type of buffer, the best way is always to check the pH of working solution, not the stock. Especially when you add some organic solvent. In this case you will avoid any potential problems.
