Can anyone give simple protocol to extract metabolites from bacterial pellets (intracellular). We tried with SDS & CTAB cell lysis method but both SDS & CTAB gives zone when we used as a control.
It all depends on what kind of bacteria you are trying to lyse and the kind of metabolite and with what activity. If you are going for antimicrobial activity, SDS and CTAB are surfactants and will definitely give zones in controls. working on metabolites, you have to avoid chemical addition for lysis. You have to go for mechanical shearing like freez thaw or bead beater techniques if sonication is not working. If you are looking for a metabolite that is heat resistant, heat can be applied... again it all comes down to the kind of metabolite being sought after.
