Hi everyone, can anyone please tell me if we zero the blank in spectrophotometer in DPPH assay? or we record the number of blank and continue reading the number for samples without doing zero for blank? Many thanks
When I mentioned blank, I mean DPPH solution whitout sample. Because I know that the more change in the color of the solution, the more antioxidant potential of the samples, so we need to measure the difference of number for DPPH solution and the sample, therefore, I wonder if we zero the dpph solution first and then measure the samples? Or do we measure the DPPH solution (without zero it) and measure sample and at the end deduct them dividing by number which spectrophotometer shows for DPPH solution? Many thanks
In fact if you zero the blank each tome or read the blank against distilled water then subtract the reading of the blank from the total Absorbance ( sample + Blank) should give you a similar result. But U should do this operation each time in both cases to eliminate any changes during individual readings. Zeroing the blank also means subtracting it reading from its presence with the sample. Same operation when you measure the blank reading against D.water the you yourself read its absorbance. Thank you and good luck
