I do MC4-L2 cell culture which is at ~100% or more of cell confluence. If I extract them and want to inject to mice, could they have their efficiency in tumor generation? Are they suitable for tumor production? Is this confluence effect on cell lines performance?
I usually use cells with 80-90% confluence for tumor injection in mice. You want cells to be healthy and in growing condition. If your cells reach 100% or more confluence, cells may be entered resting or dying stage. Since mice are expensive and tumor production may take weeks and month, I would recommend you to use cells in 80-90% confluence for mice injection.

If you are at 100% confluency you have passed exponential and likely in stationary. This means your cells are not optimal for injection and not optimal for growth in mice. You need to split your cells and harvest when you reach 85-95% confluency. If you split your flasks and grow to proper confluency then 10^6 cells incoulated BALB/c will grow tumors. Look up Lanari, et al. Cancer Research 2001, for more details.
