Dear scientist, what final concentration of DMSO should be used for 3T3-L1 cells? Some people say 0.1% while some reported 0.5 %? Will it be much more toxic if I use 0.5 % DMSO as a vehicle for my compound of interest to treat the cells?
Hi Bilal, currently I'm doing a study with 3T3-L1 cells. As far as I know, the final concentration 0.1% should be considered when using DMSO as a vehicle in 3T3-L1 cells.
The previous studies revealed that the 3T3-L1 cells is sensitive to 1% (and more) DMSO and it has cell-viability decreasing effect to the cells.
However, in my opinion, it depends on what would you do (assess) in your study. It's just my two cents but if you're going to use 0.5% DMSO, it will be much better to confirm whether there is a significant difference on the parameter that you want to assess, before you treat the cells with your compound.
0.1% is a well known concentration of DMSO to use. Because 3T3 cells are usually fast growing and robust, therefore slightly higher concentration like 0.5% can also be used.
