We did a cytotoxicity assay of 2 products using 3T3 cell line and neutral red as endpoint. We had previous IC50 data of these products in the same cell line, but with MTT. Although didn't expect it, we found big differences in the values obtained.
I thought that there should not be that difference, but has anyone had this kind of results before?

are you aware know what the cellular basis is for neutral red and MTT measurements? (answer: lysosomal staining and intracellular dehydrogenase activity, respectively) Both may be differentialy affected, depending also on the time of incubation with the drug. Do you also count cells directly?
