I passaged my ipsc's and found that the plate had debris.. so next day i changed the media but it was still there and moreover next day the debris covered the whole area of culture plate... i tried gently washing it with DPBS but couldnt' remove it... i cant understand the reason behind and how to solve it...

how thick is your plate? Your "debris" almost looks to me like the image you are getting from focusing through the plastic material. It is hard to guess from the picture you uploaded. Do you get this in the same focus layer as your cells or underneath? Is that your max magnification? Can you increase the resolution a bit more to sharpen it?

Sir the plates i have used are the standard ones manufactured by corning. No Sir I am sure about the pics, its not plastic material that I have focused on. The image is from the same focus layer as of cells.
could also be yeast but hard to tell from the images. Is the medium getting cloudy or turining pH quickly? If possible, best way to go is to trash the culture and start from scratch. Not worth the effort. Also, consider testing your stock for mycoplasma contamination.

Yes after two days of intense debris in the plate... It was cloudy beneath... N yes cells have finally died. So would be starting again but not before I figure out the reason behind... But thank you very much sir for ur support and suggestion
