What happen that caused these white spots/build up?
Just some background if needed, I cast my own gels and I ran two 15% SDS Page gels today. One of them came out perfect while my second gel came out messy. I am not new at troubleshooting my protocol but I am quite confused on what this could be.
I did cast this gel two days before use, stored inside a box with water in the 4c fridge, but I cannot see how that would effect the gel in this capacity. I appreciate any feedback!
I see a protein gel stained with silver, whose sample lanes show a ton of protein at both the high and low MW. In fact, they stain white, as it typical of silver staining. In my view, lanes are overloaded with protein of high / low MW.
Oscar L. Sierra , thank you for your answer. I did run my sample right after concentrating (I usually run a BCA assay prior to loading) so this could be the case. My protein of choice should also contain an 8kDa subunit, the final band in the ladder is 10kDa. Would it be fair to say that those build ups did not allow for the subunit to pass through or am I just making up excuses for not seeing the 8kDa band? Thank you again!
