I wish to edit a portion of a gene in a cell line using HDR. The closest PAM site (TGG) on the 5'->3' strand from databases is > 10 bp from the desired sequence i wish to edit, which I've been told isn't great. However, the opposite strand has a PAM site (AGG) that is just a bit closer. I'm wondering if I should instead design my Cas9 cut there. Does having the PAM cut before the edit site matter? The replacement oligo will span many bases before and after my edit anyways.
