I tried working CRISPR activation system to overexpress a gene which is normally not expressed in the target cell. but I failed it. I delivered each plasmids using aav (2wks infect) and already checked the system actually work in another cell type that usually express the gene.

what I can think of is that it take more time to overexpress a silent gene than the other normal expressing gene in the cell. but I am not sure about that.
