Could I transfer my cell culture I've already seeding in 12 well plate and treaded with some reagents in CO2 incubator, into 96 well plate to read the results by plat reader which couldn't read 12 well plate ??

Each well of a 12-well plate holds 1-2ml of culture medium. After having treated the cells with some reagents in a 12-well plate, the absorbance of the entire content of the well will have to be recorded. Now each well in a 96-well plate can hold a max of 250-300ul of culture medium. It won't be right to record the absorbance of only a part (250-300ul) of the entire content present in each well of the 12-well plate. It would result in erroneous readings.
The right way would be to seed cells in a 96-well plate again and treat the cells with the reagents in a reduced volume and then record the absorbance of the entire content of the well using the plate reader. It will give you proper readings.
