I am culturing cortical neurons from E15.5 mice on PDL and laminin coated plates. I plate them in Neurobasal medium supplemented with FBS, 0.6 mM L-Glut and Pen-strep. After 24 hr, I switch them to maintenance medium: Neurobasal medium with B27 supplement and pen/strep. Typically the cells in culture stay good for ~ 7 days but start dying after that. I wonder if it's because of the lack of L-Glutamine in the medium. But we have followed this protocol in a lab for a long time. Any suggestion/information on whether or not to have L-Glut and the reason behind would help.
