Hi Jawad, I guess you want to run PCR reaction and what you mentioned are initial (stock) concentrations? If yes, what is the final concentration of each? What is the final volume of your reaction? Please, provide me these information so I can help you.
if you have the original concentrations of your reagents, then you need to decide the final volume of your reaction. I usually run 20μl reactions when I am setting things up, and once my conditions are set I scale my reactions to 50μl.
To calculate the volumes for each reagent I use the following formula:
C1xV1=C2xV2
Where
C1 is the concentration of your stocks
V1 is the unknown volume you need to calculate
C2 is the concentration in the reaction
V2 is the final volume of your reaction.
I also use molecular gra water to reach the final volume.
The only thing that you really need to be careful about is the unites.
