what is the concentration of H2O2 can be use in bacterial catalase enzyme study ?
As per of observation few articles mention the direct use if H2O2 from 30% Stock, but few articles has carried out in % and as well as M concentration.
So, how to decide the concentration of hydrogen peroxide for assay in bacterial crude enzyme extract.

The concentration of H2O2 you may use in the assay depends on what kind of detection you intend to use. Do you intend to measure the direct consumption of H2O2 at 240nm? If so the initial level of absorbance has to be within the linearity domain of you spec (usually up to an OD of 2). Considering a specific coefficient of 40 M-1.cm-1 at 240nm for H2O2 the higher limit of concentration is 2/40 = 0.05 M for H2O2.
For your information, 30% (w/w) H2O2 stock solution is 9.8 M.

6% H2O2 should be OK, you can do this by applying one drop of this solution on a colony, then observe reaction; formation of bubbles means positive reaction.
