I will be working with two types of colon cancer stem cells and corresponding non-cancer stem cells. Preferably, we would like them to exploit either glycolysis or oxidative phosphorylation to a high level. For example, a type of colon non-stem cancer cell that uses mainly glycolysis and the corresponding cancer stem cells that mainly exploit oxidative phosphorylation (or vice versa). Any ideas please? Highly appreciated.

You can culture cancer stem cell-like cells from the adherent parent cell lines of the colon cancer by enrichment culturing. You can culture the adherent colon cancer cells in cancer stem cells specific media which are serum-free and containing growth factors such as B27, BFGF, EGF, and LIF at a specific concentration. The growth factor and serum-free condition will support the CSC sphere formation. Then, you need to characterize them for specific Colon cancer stem cell markers. While the respective monolayer parent cell line could be non-CSC set.
Such culturing system is widely used and established difference can be experienced since CSC are very robust to target, and this is the reason why cancer relapse or recurrence happening.
We do with breast cancer stem-like cells culturing in our lab.
However, now some company also offers cancer stem cells lines. I don't have much idea about it.

Dear Amirata Saei,
as Saurabh Mandal mentioned, you can sort out cancer stem cells from a cancer cell line, using sphere-forming assay. after that, you should sort out the CSCs by flow cytometry. for example, in our group, cancer stem cells from HCT-116 (a colon cancer cell line) was sorted out by flow cytometry, using CD133 and CD44 biomarkers.
