Hi, How do you normalize your cell numbers in viability assay to the zero hour reading
if we start with different number of cells, because after seeding cells in 96 hr, there are different cell numbers in each well of the 96 wells plate after 24 hours and before the treatment.

I have worked in cell viability. I advise you use annexin and PI by flow cytometry. Please check this link.

I am counting the growth inhibition according to the formula: Gi=[(Ai-A0)/(Ak-A0)]*100, where: : Gi — growth inhibition, A — average values of absorbance: control at the start of experiments (A0), and after the time of incubation without (Ak ) and with (Ai) the investigated compound. I am using this formula to asses growth inhibition in cancer treatment with chemotherapeutics, measured by MTT assay. Good luck!
you can also build a standard curve with known concentration of same cells to plate right before your reading (let's say 24h) and normalize your reading on that. good luck
