Can you suggest me better DNA isolation method from Plant leaf (Phyllanthus). These species have much phenols, alkaloids, mucilage ...etc. I tried CTAB, SDS, MN kit, QIAGEN kit and Charcoal-CTAB method. Still I am not getting pure DNA from some species of Phyllanthus and PCR is not working. Kindly suggest me Best, Raja
That does sound like a challenging starting material. Have you tried an "old-school" phenol/chloroform extraction? I used it to get DNA out of oily seeds with great success.
Also, you could try diluting out your DNA samples (1:2, 1:10, 1:100) and seeing if you get amplification from a diluted template. You most likely have a lot of PCR inhibiting compounds in your DNA samples. PCR is sensitive enough that less DNA is often just fine.
