I want to perform assays with cancer cell lines during the next weeks and I will grow my cells with and without FCS, afterwards I want to test different growth enhancers and growth inhibitors. Do you have some experience with that and may give me some advice which compounds are suitable?
You can try different growth factor combinations. The classic combination for cancer cells would be EGF/bFGG. I tried them in combination with PDGF and LIF, either one in combination with EGF/bFGF, or both. all combinations enhanced cell growth in different cancer cell lines, though i have to say i observed the strongest growth enhancing effect when i grew gliblastoma cells under sphere conditions without the supplementation of FBS.
The factor that stimulate or supress cell growth may depend on the type of cell and other elements, such as the surroundings, or what professor Sánchez-Fayos, from FJD, Madrid, Spain, called around 1978: 'microenvironment'. The answer to this could require all our knowledge in molecular biology, that is not my field, I'm a clinician, and have elements yet undiscovered.
Making a reduction in the scope of question, stratifying, a restriction, may help you, and also those who read the answers.
