I am looking to make an egg yolk supplement in-lab to promote the growth of Mycobacterium avium paratuberculosis. I have a method (http://vdi.sagepub.com/content/17/6/554.long) but there is no mention of sterilization. I am assuming you can't autoclave it because it would cook, but is it possible to filter sterilize it?
Egg contents are sterile when eggs are laid. So if you work properly, there should be no need to sterilize the egg yolk.
C.A. Kan, yes that's what I thought however there's always a chance of contamination if the chicken wasn't healthy. Thank you for replying!
We used eggs from healthy hens . Eggs are submerged in alcohol 70%, 30 min before use them for culture media to reduce the eggshell bacteria.

We wash the eggs first with Autoclaved or RO water to remove the visible dirt and then keep it in isopropanol for 1 hour. It works quite good.
