Since the function of both paraformaldehyde and methanol is "fixation", can we replace methanol with paraformaldehyde in the traditional gel staining solution used after SDS-PAGE?
Anand, I din´t use an aldehyde in that formula. What I used instead of methanol was isopropanolol, in the same amount, that is another alcohol. If you don´t have methanol you could try with another alcohol. Good luck!
Hi. I think that methanol is not the compound that fixed proteins in the gels. Methanol is included in buffers for transfer to blotting membranes and therefore would not be good fixative reagent. What if fixed proteins is acetic acid which then must be eliminated if what is done in silver staining proccedure. If you make Coomassie there is no problem if you use acid.
