Has anyone used immortalized cells for doing experiments and do we have to take care of any specific things while doing/performing experiments with the immortalized cells?
Any T-25 flask should work (we use corning T-25 flasks for a wide range of cell types) unless their is a specific surface your cells need to be cultured on, what cell line are you using? We use immortalized cells in lots of experiments, they don't require any specific culture conditions but again this will depend on your cell type
T25 should work fine. Just ensure you don't add too little or too much media as both will affect cell growth. When changing the media, add it to the opposite side of the flask that the cells are growing in so you don't disrupt them although in my experience immortalised cells adhere very well to T25's.
Thank you everyone for the suggestions and opinions. For the better understanding of the question we are using immortalized HUVEC cells. Does this type of cells requires any kind of special coating on the T-25 flask?
Hi I also do not have any problem in culturing cells in normal flasks. At the moment my cells are Vero, WRL-68 and 3T3 cells. They are growing well in normal flasks for my in-vitro experiments. If the cells are not growing well, I found it is mainly due to contamination and not by the materials from the flasks.
As all above have said also, I don't see why you can't use the T-25 flasks. We use these to culture a variety of immortalised human and fish cell lines without any issues.
Since the source of your cells is recommending a specific flask I would experiment by using that flask and another plain flask and see if you notice any differences in the way they attach using the type of media they recommend. Not familiar with the specific cells you refer too, but some immortalized cell lines do need a matrix in order to maintain certain functionality.
I have been using Caco-2 and MDCK-MDR1 cell line for many years using non-coating T25, T75 flasks without any problem. Usually, the immortalized cell culture does not need special coating for the flasks used except special cell line like neuron cell. However, the types of medium used are very important. You have to prepare medium carefully according to handling manual for the cell line.
If you would not like to waste time in experimenting with the T-25 flasks, then use the ones specified by the provider. This is so, because you will have more important experiments to do with immortaliuzed cells, rather than experimenting with T-25 flasks.
Yes. T25 flasks should work from my experience. Collagen coating helps the cells grow too. I use Cat. G422 (https://www.abmgood.com/Applied-Cell-Extracellular-Matrix-G422.html) from ABM.
