I always make fresh glycerol stocks from the primary culture and use them for 2-3 months, before subculturing and making another batch and store it at-80. I have been doing this for 4-5 times. All of a sudden, I notice during purification that the yield is reduced quite significantly. I suspect this could be due to lower level of expression and on checking the newly made stocks (I did my purification from this one's parent stock) I found there was NO expression. I also did fresh transformation and got colonies in my kanamycin plate but even they do not show overexpression at all. Is there something wrong with my clone? I have tried picking several colonies from that plate and I am currently checking them as well. Also I had some old stock which also I am checking. I also checked whether Bl-21De3 cells (I am using them) had somehow become resistant to kanamycin (Ab i use), but on streaking the cells on a freshly made kan plate, there were no colonies, so false positive colonies are essentially ruled out. Please suggest something. And also why could this possibly happen?
