Sub-micrometer particles are usually measured by electron microscopy or by dynamic light scattering. Can a flow cytometer be used instead to distinguish between particle populations below 1 micrometer? For example can it be used to see that there are three distinct particle groups - 100 nm, 200 nm and 500 nm?
Ok, I just contacted a Becman Coulter specialist with this question and here is what they said - for the benefit of those who might be interested:
If the particles are fluorescent they can be measured by any flow cytometer. If they are not a cytometer with a blue laser will be able to measure with 'scatter' particles larger than 400 nm and if you have a violet laser (405 nm wavelength) you can measure above 80 nm.
NTA (nanotracking analysis) is another option in this size range. And NTA can be used to see that there are three distinct particle groups - 100 nm, 200 nm and 500 nm

the answer of Beckman Coulter answers the question at least in part. My impression is that a violet laser in theory can see different populations of particle with size above 80 nm. It is a completely different question if the particles will have discrete populations or will be spread in size across a range. If you see it differently I will be grateful if you make a statement and not just post another question.

we have since years flow cytometers in my lab but we use them only with cell so far. I think we can say safely that we know the principles. The question was on specific future application with which we have no experience whatsoever.
