We would like to simply put a bright EGFP plasmid in human primary cells with the purpose of tracking and evaluating transfection efficiency. The plasmid mEGFP-Lifeact-7 looks suitable to me:

However I don't know if the cytomegalovirus enhancer-promoter will work well in primary human cells and I will be grateful if anybody with knowledge and experience can answer this question.

Which primary cell type were you planning to use? I have previously expressed CMV driven maxGFP quite successfully in human primary T-lymphocytes via nucleofection. Therefore, I know CMV promoters work at least in some human primary cell types. Only concern however is possible cell-specific silencing of CMV promoter. Studies have shown that this can happen in certain cell types. So, I guess you would have to try out and find whether it is the case in cells that you plan to use. Hope this helps and good luck!
