I have extracted steroids using three methods: ether, ethanol:acetone and sonication in hypotonic buffer followed by high speed centrifugation at 50000 g for 30 min. The recovery was different for all three procedures, highest being in third method followed very closely by ethanol:acetone method followed by ether. Both second and third methods had very close recoveries. But do these methods help extract total steroids? Both cytoplasmic and nuclear. And also the steroids bound to their high affinity and low affinity receptors? I could not get resources which specifically mention this.
