I am looking for an anti-dsDNA antibody. (Could also be an anti-ssDNA antibody). Can someone suggest a good and specific one? I would like to precipitate sheared Protein-chromatin complexes with it.
Your question is a bit obscure. Do you want to use an anti-dsDNA assay to screen sera in order to find a positive source that can be used for immunoprecitation? And why would anti-ssDNA be OK for this purpose?
There are several good ways to do anti-dsDNA, but they depend on some infrastructure. If you can do ELISA, there is a protocol I could send you. If you have access to a fluorescence microscope and some money, you can screen on Crithidia slides. But you need to clarify your comment about anti-ssDNA in order to decide which method would be appropriate.
Dear Robert, thank you for your answer! Sorry for the lack of clarity. I actually do not want to perform an anti-dsDNA assay. I don´t want to check for anti-dsDNA antibodies in serum. I would like to perform chromatin immunoprecipitation. My formaldehyde-crosslinked cell lysate is sheared by sonication. Then I would like to apply the anti-dsDNA antibody to precipitate sheared chromatin. Afterwards I would like to check by WB how much of my protein of interest is bound to DNA, in comparison to my control sample. (I am interested in non-specific DNA binding of a transcription factor).
