I would like to transport for the anaerobic chamber to a plate reader outside. I'm looking at various seals etc. to make the plate anaerobic but any guidance would be appreciated.
Hi Ciara, are you asking how you can "make" it or "keep" it anaerobic. If the former, I imagine you are referring to an approach to get any O2 out of the plastic. Maybe you can keep it in the chamber for a few days?
Thank you, I think this would be a good idea and some people in the lab have suggested to use a lighter to melt the edges of the parafilm in order to seal fully. Of course this would have to be done outside the chamber.

I mean both actually. I plan on keeping plates inside the chamber before attempted use as it would take a few days to remove oxygen from the plastic. But then to bring to the plate reader I had thought to seal with a film and maybe silicone, while also placing degassed mineral oil on top of each well. Some discussions that have came up were to cover in parafilm and melt with a lighter. Dr. Jan Arends has also had some discussions about this (I'm waiting for an email back) but he seals the corner wells with vaseline also. I'm curious to see will it work!
