I wuold like to apply the acetolysis method described by Erdtman(1960) in pollen samples. Is it possible to use as sample already glycerin pollen slides for L.M.?
I think your question is: can I use acetolysis on pollen that are already mounted in glycerin for LM. In that case: I think you can! Just heat the microscopic slide on a heating plate to open it up, scrape off the glycerin containing the pollen grains and then use that as your input for the acetolysis protocol. Be sure to work in a fume hood and use protective wear, since depending on the water content of the sample there could be some reaction. This may be something to consider to be careful, see e.g. Jones et al, 2014 (Pollen analyses for pollination research, acetolysis).
