I want to do CRISPR and ShRNA for few genes and I came a cross few studies that knocked-out those genes and they shared in the method sections the sequences that worked. usually for CRISPR and ShRNA we use multiple sgRNA and siRNA ,respectively to knock-out the gene. My question is : do I need to do so even if those studies shared to sequences that worked with them? or there is no guarenatee that it will work for me?

And if that is the case, how many siRNA and sgRNA should I use ?

and the other thing is that is it a must to have search or motifs such as 5′-AA(N19)TT, 5′-AA(N21) or 5′-NA(N21) on the target sequence?
