I want to use ORI in my plasmid vector for replicating it in SH-SY5Y cells like in 293E or 293T cells with SV40?
Is there any ORI that can be replicated in SY5Y cells?
Interesting question. Following the discussion.
Mammalian replication origins are poorly defined, and usually consist of large regions. There is no equivalent to yeast ARS sequences or bacterial Oris that can be inserted into a plasmid and allow it to be replicated.
The SV40 ori/T-antigen is the exception.
I am not sure; is AAV2 best for SY5Y and HT22? Is there any comparative study for these lines? Also, Can i use same serotype vector for the two cell lines?
