I want to do immunostaining of the corneal endothelial cells of sheep origin. Just wondering that what are the alternatives of goat serum as blocking buffer, i thought BSA or FCS.
as Duy Q Phan mentioned you should first try serum from the secondary antibody species!
If this is not possible, it is not allowed or recommended to use serum or serum albumines from the primary antibody species (due to cross reactions)!
So, if you do not use cow primary antibodies, BSA or calf serum (is cheaper than fetal calf serum) can be used. I use it for blocking steps in a concentration of 3-5% and I add it to primary and secondary antibodies in a concentration from 1-3% (low amounts if i add also serum, and higher amounts if BSA is used alone).
