In an order to check for DNA protection on X-ray/gamma irradiated DNA samples treated with any radioprotective drug, plasmid relaxation assay is often been cited as an easy tool, where plasmids of known sizes like pBR322 are commonly used. Although the assay is specific for plasmids as it is based upon the percentage of appearance of the supercoiled and open circular forms which is not possible in genomic DNA. Is there a possible way to do the same assay with genomic DNA of any other origin for checking DNA protection with radioprotective drugs on the basis of appearance of bands on agarose gel.
The experiment is strictly based on naive DNA only without any cell system so the question of COMET assay etc., is totally ruled out for me.
