Hello, i did a cell viability assay on a cell line after exposure to a test substance.

The test worked fine and was valid in all criteria.

The cells were stained with alamar blue and CFDA-AM as well as Neutral Red.

The cells showed 40% viability for AlamarBlue for the highest test substance concentration, but 100% viability for CFDA-AM. For Neutral Red, viability was 70%.

Since the test was valid and all controls looked fine, i dont think it is a contamination. Because if it was, the signal in the cell free control would be higher...

Now my question would be: Is it possible, that the test substance disturbed the metabolism (AlamarBlue) but not the cell membrane integrety?

Does someone have any suggestion how this could be possible (maybe with a tipp for papers)?
